Seems Simple Backend is not yet enabled by default in Chromium 54.
So it is not necessary to change that flag.
Can you add the command line to check the result?
--force-fieldtrials=SimpleCacheTrial/Disabled/

Another possibility is cache limit reached.
You can try adding this command line to chrome.exe:
--disk-cache-size=5368709120
